Endothelial Seeding as an Method to Improve Patency Rates of Denuded Vascular Surfaces (An Experimental Study)

Endothelial seeding is assumed to be an effective
method to improve patency rates of denuded vascular surfaces that is seen after angioplasty. In this
study, following anesthesia jugulary veins of eight dogs where harvested bilaterally. From these veins
endothelial cells were obtained enzymatically by
collagenase and were passaged in to culture mediums until they reach a satisfactory number. After
three weeks, dogs were anesthesied similar fashion.
Their femoral veins were exposed and experimental
intimal denudation were performed bilaterally. Subsequently, one of femoral artery was injected with
cell solution and the other with saline solution. Two
weeks after injections, arteriographic studies of femoral arteries were performed and arterial specimens were taken for histological evaluation. We concluded
that patency rates were statistically significant between study and control sites (p<0.05). Histologic evaluations has also revealed that cultured endothelial cells have attached to the damaged surfaces.
There were no cells on the control site. Thrombüs
formation was also seen on the damaged subintimal
layer despite there was nothing on the study site.
